Abstract
Transdermal drug delivery offers several advantages over conventional oral and parenteral administration; however, the stratum corneum presents a major barrier to the effective delivery of many therapeutic agents. Rapid-dissolving microneedles have emerged as a minimally invasive approach capable of creating temporary microchannels in the skin and facilitating drug transport. Among advanced nanocarriers, Nano-Transfersomes are particularly attractive because of their high deformability, nanoscale size, and ability to enhance drug permeation across biological barriers. The integration of nano-transfersomes into rapidly dissolving microneedle patches combines the penetration-enhancing capability of microneedles with the drug-delivery advantages of flexible lipid vesicles. This review discusses the principles, classification, materials, fabrication techniques, mechanisms of skin penetration and drug release, and therapeutic applications of rapid-dissolving microneedle patches loaded with nano-transfersomes. Particular emphasis is placed on their potential applications in osteoporosis, cancer, vaccination, infectious diseases, dermatological disorders, and delivery of biological therapeutics. Recent advances, formulation challenges, stability considerations, manufacturing approaches, and future opportunities for personalized and targeted transdermal therapy are also discussed. Overall, nano-transfersome-loaded rapid-dissolving microneedles represent a promising next-generation platform for improving transdermal drug delivery and may facilitate the development of safer, more effective, and patient-friendly therapeutic systems.
